This antibody recognizes a protein of 25-26 kDa, identified as Bcl-2. This antibody shows no cross-reaction with Bcl-x or Bax proteins. Bcl-2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. Bcl-2 appears to function in a feedback loop system with caspases. It inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1). Expression of Bcl-2 inhibits programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of Bcl-2 protein, whereas the normal or hyperplastic germinal centers are negative. Recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO) (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Biotium CD66 (CEA)(COL-1), CF568 conjugate, 0.1mg/mL
This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA positive) from pleural mesotheliomas (rarely or weakly CEA positive). Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. This MAb selectively interferes with lymphocyte binding to lymph node, mucosal and synovial endothelium. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. This antibody reacts with Ig domain of CD47 protein. It has been shown to inhibit polymorphonuclear neutrophil (PMN) transmigration across cell monolayers and matrix. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. This antibody recognizes a protein of 26 kDa, identified as CD81 (Workshop VI; Code CD81.1). CD81 has a very broad cellular distribution, being expressed on T- and B-lymphocytes, NK cells, thymocytes, eosinophils, fibroblasts, epithelial and endothelial cells. Neutrophils, erythrocytes and platelets are negative, while monocytes are variably positive. CD81 is a member of a family of tetraspanin transmembrane proteins, including CD9, CD37, CD53, CD63, and CD82. It associates with CD19, CD21, Leu 13, and integrins on cell membrane and is involved in signal transduction in B lymphocyte development and cell adhesion. CD81 also acts as a receptor for the envelope protein E2 of chronic hepatitis C virus.
